=== mEos4 === Many new monomeric versions of EosFP have been developed that offer advantages over wild type EosFP. Developed by a team at the Janelia Farm Research Campus at Howard Hughes Medical Institute, mEos4 has higher photostability and longer imaging abilities than EosFP. It is also highly resistant to chemical fixatives such as PFA, gluteraldehyde and OsO4 which are used to preserve samples. mEos4 is effective at higher temperatures than EosFP, phot-converts at an increased rate and has a higher emission amplitude in both green and red fluorescent states. Applications for the mEos4 protein include photoactivation localization microscopy (PALM), correlative light/ electron microscopy (CLEM), protein activity indication and activity integration (post-hoc imaging for protein activity over time).

Pharmaceutical medications are commonly described as "ubiquitous" in nearly every type of environmental medium (i.e. lakes, rivers, streams, estuaries, seawater, and soil) worldwide. Scientists generally divide environmental impacts of a chemical into three primary categories: persistence, bioaccumulation, and toxicity. Since medications are inherently bio-active, most are naturally degradable in the environment, however they are classified as "pseudopersistent" because they are constantly being replenished from their sources. These Environmentally Persistent Pharmaceutical Pollutants (EPPPs) rarely reach toxic concentrations in the environment, however they have been known to bioaccumulate in some species. The T5 exonuclease chews back DNA from the 5' end of each fragment, exposing 3' overhangs on each DNA fragment. The complementary overhangs on adjacent DNA fragments anneal via complementary base pairing. The Phusion DNA polymerase fills in any gaps where the fragments anneal. Taq DNA ligase repairs the nicks on both DNA strands. Because the T5 exonuclease is heat labile, it is inactivated at 50 °C after the initial chew back step. The product is thus stable, and the fragments assembled in the desired order. This one-pot protocol can assemble up to 5 different fragments accurately, while several commercial providers have kits to accurately assemble up to 15 different fragments in a two-step reaction. However, while the Gibson assembly protocol is fast and uses relatively few reagents, it requires bespoke DNA synthesis as each fragment has to be designed to contain overlapping sequences with the adjacent fragments and amplified via PCR. This reliance on PCR may also affect the fidelity of the reaction when long fragments, fragments with high GC content or repeat sequences are used.

The yellow pulp, which contains the seeds, is naturally sweet and is processed into a valuable carbohydrate food. The seeds are processed into a seasoning, known as sikomu and dodowa/ dawadawa among the Yoruba and Hausa people respectively, and sunbala in the Inner Niger Delta. The pulp is also used to make beverages. The seeds of the locust bean are the most valuable part of the plant. They are high in lipids (29%), protein (35%), carbohydrates (16%), and are a good source of fat and calcium for rural dwellers. The seed is first cooked to remove the seed coat and then fermented to produce the desired result. Seeds can then be pounded into powder, then formed into cakes. In a study conducted on the fermentation of dawadawa, it was found that Gmelina arborea and banana leaves accelerated fermentation of seeds, while increasing fat, protein and moisture content, but also decreasing carbohydrate content.

Negrin opened wide, fascinating vistas to my imagination, not only through his lectures and laboratory teaching, but through his advice, encouragement, and stimulation to read scientific monographs and textbooks in languages other than Spanish. Negrín encouraged Ochoa and another student, José Valdecasas, to isolate creatinine from urine. The two students succeeded and also developed a method to measure small levels of muscle creatinine. Ochoa spent the summer of 1927 at University of Glasgow working with D. Noel Paton on creatine metabolism and improving his English skills. He also refined the assay procedure further and upon returning to Spain he and Valdecasas submitted a paper describing the work to the Journal of Biological Chemistry, where it was rapidly accepted, marking the beginning of Ochoa's biochemistry career. Ochoa completed his undergraduate medical degree in the summer of 1929 and decide to go abroad again to gain further research experience. His creatine and creatinine work led to an invitation to join Otto Meyerhof's laboratory at the Kaiser Wilhelm Institute for Biology in Berlin-Dahlem in 1929. At that time the institute was a "hot bed" of the rapidly evolving discipline of biochemistry, and thus Ochoa had the experience of meeting and interacting with scientists such as Otto Heinrich Warburg, Carl Neuberg, Einar Lundsgaard, and Fritz Lipmann in addition to Meyerhof who had received the Nobel Prize in Physiology and Medicine less than a decade earlier.
