Glutathione is a small sulfur-containing peptide built from glutamic acid, cysteine, and glycine. Its distinctive feature is a gamma-glutamyl bond between glutamate's side-chain carboxyl group and cysteine's amino group. This linkage resists ordinary peptidases and helps the molecule remain stable inside cells. The reduced thiol form, often abbreviated GSH, is the dominant intracellular species. The oxidized disulfide dimer, GSSG, forms when two reduced molecules link through their cysteine sulfur atoms. The balance between these forms is a common redox indicator.
In living systems, glutathione occurs in millimolar concentrations in many cell types, while extracellular levels are generally much lower. The liver holds a substantial share of the body's total pool, and the molecule participates in reduction, detoxification, and amino acid transport. It also serves as a cofactor for enzymes such as glutathione peroxidase and glutathione S-transferase. Because the cysteine residue supplies a reactive thiol, glutathione can donate electrons and become oxidized. Cells regenerate reduced glutathione through glutathione reductase using NADPH.
Enzymatic recycling assays provide a complementary approach for total glutathione. In these methods, glutathione reductase reduces oxidized glutathione while a thiol-reactive reagent, such as 5,5'-dithiobis(2-nitrobenzoic acid), produces a colored product. The reaction cycles between reduced and oxidized forms, amplifying the signal. Spectrophotometric or fluorometric detection can then estimate concentration. Distinguishing reduced glutathione from glutathione disulfide often requires separate aliquots, masking agents, or chromatographic separation, and the choice affects reported values.
Storage conditions strongly influence glutathione stability. The solid reduced form is commonly kept desiccated at or below minus twenty degrees Celsius, protected from light and moisture. Aqueous solutions are less stable because the thiol group reacts with dissolved oxygen, and oxidation accelerates at neutral or alkaline pH. Acidic solutions and oxygen-free handling can slow degradation, but repeated freeze-thaw cycles should be avoided. Researchers often verify concentration before use, because apparent losses can arise from oxidation or water uptake.

The glass bulb of a general service lamp can reach temperatures between 200 and 260 °C (392 and 500 °F). Lamps intended for high power operation or used for heating purposes will have envelopes made of hard glass or fused quartz. If a light bulb envelope leaks, the hot tungsten filament reacts with air, yielding an aerosol of brown tungsten nitride, brown tungsten dioxide, violet-blue tungsten pentoxide, and yellow tungsten trioxide that then gets deposited on the nearby surfaces or the bulb interior.

== Effects of MPPT on wound healing == MPPT has been evaluated in a preclincial wound healing model and in a 266 patient comparative clinical study, which included a wide range of wound types. The study found that MPPT reduced the time to achieving an infection-free and healing wound by 60% compared to a topical antibiotic (genamicin) and to the antiseptic iodine. MPPT also reduced the number of hospitalisation days by 31% compared to the antibiotic. At Bristol University Hospital, MPPT was evaluated for use on wounds. The study included nine dehisced surgical wounds and one category 4 pressure ulcer. Standard-of-care for these types of wounds are one week with UrgoClean followed by 2 or more weeks with Negative Pressure Wound Therapy (NPWT). Wounds receiving MPPT were able to achieve the same stage of wound healing in 4–5 days as would have required 3 or more weeks with standard-of-care, thus offering savings of 67%. All wounds receiving MPPT closed. MPPT has also been used on diabetic foot ulcers and venous leg ulcers, including ulcers that were chronic and non-healing and in all cases it has been able to promote healing. MPPT has also been used on pressure ulcers, including chronic non-healing ulcers and was in a similar manner able to promote healing. A poster was recently presented by the Birmingham University Hospitals at the British Association of Dermatologists, which showed that MPPT was able to assist the healing of 3 chronic, stable pyoderma gangrenosum ulcers. In one patient, it was possible to reduce the dose of immunosuppressant.

internal validation or cross-validation (actually, while extracting data, cross validation is a measure of model robustness, the more a model is robust (higher q2) the less data extraction perturb the original model); external validation by splitting the available data set into training set for model development and prediction set for model predictivity check; blind external validation by application of model on new external data and data randomization or Y-scrambling for verifying the absence of chance correlation between the response and the modeling descriptors. The success of any QSAR model depends on accuracy of the input data, selection of appropriate descriptors and statistical tools, and most importantly validation of the developed model. Validation is the process by which the reliability and relevance of a procedure are established for a specific purpose; for QSAR models validation must be mainly for robustness, prediction performances and applicability domain (AD) of the models. Some validation methodologies can be problematic. For example, leave one-out cross-validation generally leads to an overestimation of predictive capacity. Even with external validation, it is difficult to determine whether the selection of training and test sets was manipulated to maximize the predictive capacity of the model being published. Different aspects of validation of QSAR models that need attention include methods of selection of training set compounds, setting training set size and impact of variable selection for training set models for determining the quality of prediction.

Raw opium may be sold to a merchant or broker on the black market, but it usually does not travel far from the field before it is refined into morphine base, because the pungent, jelly-like form of raw opium is bulkier and harder to smuggle. Crude laboratories in the field are capable of refining opium into morphine base by a simple acid-base extraction. A sticky, brown paste, morphine base is pressed into bricks and sun-dried, and can either be smoked, prepared into other forms or processed into heroin. Other methods of preparation (besides smoking), include processing into regular opium tincture (tinctura opii), laudanum, paregoric (tinctura opii camphorata), herbal wine (e.g., vinum opii), opium powder (pulvis opii), opium sirup (sirupus opii) and opium extract (extractum opii). Vinum opii is made by combining sugar, white wine, cinnamon, and cloves. Opium syrup is made by combining 97.5 part sugar syrup with 2.5 parts opium extract. Opium extract (extractum opii) finally can be made by macerating raw opium with water. To make opium extract, 20 parts water are combined with 1 part raw opium which has been boiled for 5 minutes (the latter to ease mixing). Heroin is widely preferred because of increased potency. One study in postaddicts found heroin to be approximately 2.2 times more potent than morphine by weight with a similar duration; at these relative quantities, they could distinguish the drugs subjectively but had no preference. Heroin was also found to be twice as potent as morphine in surgical anesthesia.
== History == The US Food and Drug Administration (FDA) approved trastuzumab deruxtecan based on the results of one clinical trial enrolling 184 female participants with HER2-positive, unresectable and/or metastatic breast cancer who had received two or more prior anti-HER2 therapies in the metastatic setting. These participants were heavily pretreated in the metastatic setting, receiving between two and 17 therapies prior to receiving trastuzumab deruxtecan. Participants in the clinical trial received trastuzumab deruxtecan every three weeks and tumor imaging was obtained every six weeks. The overall response rate was 60.3%, which reflects the percentage of participants who had a certain amount of tumor shrinkage with a median duration of response of 14.8 months. Efficacy was evaluated in a multicenter, open-label, randomized trial (DESTINY-Gastric01, NCT03329690) in participants with HER2-positive locally advanced or metastatic gastric or GEJ adenocarcinoma who had progressed on at least two prior regimens, including trastuzumab, a fluoropyrimidine- and a platinum-containing chemotherapy. A total of 188 participants were randomized (2:1) to receive trastuzumab deruxtecan 6.4 mg/kg intravenously every three weeks or physician's choice of either irinotecan or paclitaxel monotherapy.
During each turn of the cycle, two carbon atoms leave the cycle as CO2 in the decarboxylation reactions catalyzed by isocitrate dehydrogenase and alpha-ketoglutarate dehydrogenase. Thus each turn of the citric acid cycle oxidizes an acetyl-CoA unit while regenerating the oxaloacetate molecule with which the acetyl-CoA had originally combined to form citric acid. The decarboxylation reactions occur before malate is formed in the cycle. Malate is the only substance that can be removed from the mitochondrion to enter the gluconeogenic pathway to form glucose or glycogen in the liver or any other tissue. There can therefore be no net conversion of fatty acids into glucose. Only plants possess the enzymes to convert acetyl-CoA into oxaloacetate from which malate can be formed to ultimately be converted to glucose.
