Glutathione is a small tripeptide made of glutamic acid, cysteine, and glycine. Its cysteine thiol group allows reversible oxidation and reduction, making it central to cellular redox chemistry. The reduced form, often abbreviated GSH, predominates inside most cells, while the oxidized disulfide form, GSSG, forms when two GSH molecules react. The ratio of GSH to GSSG is widely used as an indicator of oxidative stress in laboratory research, though it does not by itself diagnose a clinical condition.
Biosynthesis occurs in two ATP-dependent steps. The enzyme glutamate-cysteine ligase joins glutamate and cysteine, forming gamma-glutamylcysteine; glutathione synthetase then adds glycine to produce the complete tripeptide. Because the peptide bond from glutamate uses the gamma-carboxyl group, glutathione resists digestion by many ordinary peptidases. Tissues vary in synthesis capacity, and the liver generally contains high concentrations relative to many other organs. This uneven distribution contributes to organ-specific differences in redox buffering and affects how experimental results are interpreted across tissue types.
Laboratory measurement of glutathione requires attention to oxidation before analysis. Blood, tissue, or cell samples can lose reduced glutathione as it converts to GSSG or forms mixed disulfides with proteins. Acid extraction, rapid freezing, and thiol-blocking reagents are common strategies to preserve the original distribution. Reported concentrations therefore depend on collection protocol, extraction method, and the time between sampling and analysis. Comparisons across studies are most reliable when these pre-analytical variables are described.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. The enzymatic recycling assay uses glutathione reductase and a colorimetric or fluorometric reagent to amplify signal, which gives good sensitivity for total glutathione. Chromatographic methods can separate GSH from GSSG and related thiols, while mass spectrometry offers structural confirmation and multiplexing. Each approach has different requirements for calibration, internal standards, and validation. No single method captures every form of glutathione in every matrix.

Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. It contains an unusual gamma-glutamyl bond between glutamate and cysteine, which resists cleavage by many peptidases. The reduced form, GSH, carries a thiol group on cysteine and is the dominant intracellular form in most cells. Its structure allows it to participate in redox reactions and to serve as a sulfur donor. The oxidized form, GSSG, consists of two GSH molecules joined by a disulfide bond.
In cells, glutathione helps maintain the reducing environment of the cytosol and supports enzymes that counteract reactive oxygen species. It acts as a cofactor for glutathione peroxidases, which reduce hydrogen peroxide and lipid peroxides, and for glutathione S-transferases, which conjugate electrophiles. The ratio of GSH to GSSG is often used as an indicator of oxidative stress, although the ratio can vary by compartment and cell type. Glutathione also stores cysteine, an amino acid that can be limiting for protein synthesis and antioxidant defense.
Common analytical approaches include enzymatic recycling assays, high-performance liquid chromatography, and mass spectrometry. Enzymatic recycling measures total glutathione after converting GSSG back to GSH, while separation methods can quantify GSH and GSSG separately. Derivatization may be used to improve detection or stability during analysis. LC-MS/MS offers high specificity and can distinguish glutathione from related thiols and adducts. Each method has different sensitivity, throughput, and susceptibility to interference, so method selection depends on the study question and sample matrix.
For solid glutathione reagents, storage at low temperature and protection from moisture and light are typical precautions. Aqueous solutions can oxidize over time, and pH affects stability; alkaline conditions generally promote thiol oxidation. Some protocols prepare fresh solutions, while others use antioxidants or chelators to limit metal-catalyzed oxidation. Purity and counterion content can vary among commercial preparations, affecting concentration calculations. Certificates of analysis and validated assays help verify identity and purity.
Relatedly, muscimol, similarly to gaboxadol, has been described as being highly though incompletely selective for these GABAA receptors. In contrast, benzodiazepines and Z-drugs do not activate δ subunit-containing GABAA receptors. On the other hand, alcohol is known to selectively potentiate δ subunit-containing extrasynaptic GABAA receptors analogously to muscimol. While muscimol is often thought of as a selective GABAA agonist with exceptionally high affinity to δ subunit-containing GABAA receptors, it is also a potent partial agonist of the GABAA-ρ receptor, and so its range of effects results from a combined action on more than one GABAA receptor subtype. In fact, it is more potent as a partial agonist of the GABAA-ρ receptor than as a GABAA receptor agonist. Muscimol has been said to be inactive at the GABAB receptor. However, a subsequent study reported that muscimol may have GABAB receptor-mediated inhibitory activity, although more research is needed to further characterize this activity. Muscimol is inactive in terms of affecting GABA transaminase (GABA-T). There is little evidence that muscimol interacts with other biological targets besides the GABA receptors and the GABA transporters. Muscimol shows a very steep dose–response curve in rodents. It produces effects in rodents including central depression, hypolocomotion, catalepsy, sedation, ataxia, analgesia, anxiolysis, anticonvulsant effects, neuroprotective effects, and anesthesia, among other effects.
=== Muscle contraction === ATP fuels muscle contractions. Muscle contractions are regulated by signaling pathways, although different muscle types being regulated by specific pathways and stimuli based on their particular function. However, in all muscle types, contraction is performed by the proteins actin and myosin. ATP is initially bound to myosin. When ATPase hydrolyzes the bound ATP into ADP and inorganic phosphate, myosin is positioned in a way that it can bind to actin. Myosin bound by ADP and Pi forms cross-bridges with actin and the subsequent release of ADP and Pi releases energy as the power stroke. The power stroke causes actin filament to slide past the myosin filament, shortening the muscle and causing a contraction. Another ATP molecule can then bind to myosin, releasing it from actin and allowing this process to repeat.
Retaliating, Gaddafi sponsored anti-government militants in Tunisia into the 1980s. Turning his attention to Algeria, in 1975 Libya signed, in Hassi Messaoud, a defensive alliance allegedly to counter alleged "Moroccan expansionism", also funding the Polisario Front of Western Sahara in its independence struggle against Morocco. Seeking to diversify Libya's economy, Gaddafi's government began purchasing shares in major European corporations like Fiat as well as buying real estate in Malta and Italy, which would become a valuable source of income during the 1980s oil slump.

The PRIDE (PRoteomics IDEntifications database) is a public data repository of mass spectrometry-based proteomics data, and is maintained by the European Bioinformatics Institute as part of the Proteomics Team. Originally designed by Lennart Martens in 2003 during a stay at the European Bioinformatics Institute as a Marie Curie fellow of the European Commission in the "Quality of Life" Programme (Contract number: QLRI-1999-50595), PRIDE was established as a production service in 2005. The original grant application document from June 2013 to start construction of PRIDE has since been published in a viewpoint article. Several similar proteomics databases have been built, including the GPMDB, PeptideAtlas, Proteinpedia and the NCBI Peptidome. The PRIDE database constitutes a structured data repository, and stores the original experimental data from the researchers without editorial control over the submitted data. In total, PRIDE contains data from about 60 species, the biggest fraction of it coming from human samples (including the data from the two draft human proteomes) followed by the fruit fly Drosophila melanogaster and mouse.

stringency The effect of conditions such as temperature and pH upon the degree of complementarity that is required for a hybridization reaction to occur between two single-stranded nucleic acid molecules. In the most stringent conditions, only exact complements can successfully hybridize; as stringency decreases, an increasing number of mismatches can be tolerated between the two hybridizing strands.
==== Post mortem interval ==== Post mortem interval (PMI) is the time between death and discovery of a corpse. Calliphora vomitoria is important for PMI estimations because it is among the first species to lay eggs on the corpse. There are two ways of estimating PMI. One is killing the larvae, and then comparing the larvae's length and temperature to those in the standardized data. Another way to calculate PMI is to calculate the accumulated degree hours/days (ADH/D) that a larva needs to reach a certain developmental stage. The later method is the more widely accepted way to estimate PMI.
== Modern Development: Sector 13 == For decades, Manimajra sat outside Chandigarh's formal grid because the city's planner, Le Corbusier, omitted the number 13 due to superstition. Following long-standing demands from residents in upscale enclaves like the Modern Housing Complex (MHC) and Shivalik Enclave, the area was officially integrated into the sectoral grid as Sector 13 in February 2020. This change was enacted by Governor V.P. Singh Badnore under the Capital of Punjab Act of 1952.
=== Jinye === Closely related to xuě are the jinye (津液; jīnyè, usually translated as "body fluids"), and just like xuě they are considered to be yin in nature, and defined first and foremost by the functions of nurturing and moisturizing the different structures of the body. Their other functions are to harmonize yin and yang, and to help with the secretion of waste products. Jinye are ultimately extracted from food and drink, and constitute the raw material for the production of xuě; conversely, xuě can also be transformed into jinye. Their palpable manifestations are all bodily fluids: tears, sputum, saliva, gastric acid, joint fluid, sweat, urine, etc.

GSH is the reduced form of glutathione, with a free thiol group on cysteine. GSSG is the oxidized disulfide form, created when two GSH molecules become linked. The two forms exist together, and their balance is often reported as the GSH/GSSG ratio in laboratory studies.
Glutathione is synthesized inside cells from amino acids rather than being classified as an essential dietary nutrient. Dietary sources can provide glutathione or its precursors, but digestion and absorption alter what reaches tissues. Research continues on how dietary intake relates to cellular glutathione levels.
The liver has high glutathione concentrations and uses the compound in conjugation and antioxidant reactions. These reactions are relevant to the processing of drugs, pollutants, and normal metabolic byproducts. Studies often examine liver glutathione as a marker of oxidative stress or detoxification capacity.
Pre-analytical handling, extraction chemistry, and detection method all influence reported glutathione values. Oxidation during sample processing can shift the measured GSH/GSSG ratio. Standardized protocols and reference materials help reduce, but do not eliminate, these differences.
